The VersaLive master mold can be replicated in a cleanroom and with basic photolithography skills. VersaLive was conceived and designed to be fabricated in one single layer so that no head-scratching alignment steps are required.
In our experience, a 20-μm-high channel can accomodate a large variety of cell lines (e.g. HeLa, CHO, HCC38, AU565).
As general rule, however, we advise in using channels 20 -- 50% higher than the dimension of your cells when they are in suspension (round). In this way, they will be easily loaded into the chambers without forming multi-layers.
